Gilby John Clarke (født 17. august 1962, i Cleveland, Ohio) er en amerikansk gitarist som er mest kjent for sin periode i Guns N' Roses, hvor han erstattet gitarristen Izzy Stradlin i november 1991. Han spilte med bandet til 1994. Det året ble hans første soloalbum; Pawnshop Guitars, utgitt.
